Merck: New pneumococcal reference serum

With more than 100 pneumococcal serotypes in circulation, new pneumococcal conjugate vaccines containing serotypes are needed to address the changing pneumococcal serotype epidemiological landscape.  Merck & Co. developed a new pneumococcal reference standard (MPRSS-01) containing serotype-specific IgG concentrations for 24 serotypes from the existing standard (007sp), as well as with an additional nine novel serotypes.  The antibody concentrations for the 24 established serotypes in MPRSS-01 were assigned via direct calibration to the existing standard, while a cross-standardization approach was used for the nine novel serotypes.  The calibration study demonstrated parallelism of the dilution-response curves across the 33 serotypes between MPRSS-01 and 007sp, allowing the use of any 007sp-assigned serotype as a reference calibrator.
